Rôle des principales voies inflammatogènes impliquées dans les maladies inflammatoires chroniques de l’intestin dans le contrôle de l’homéostasie protéolytique intestinale.
La maladie de Crohn et la rectocolite hémorragique sont des maladies inflammatoires chroniques de l’intestin (MICI). Trois grandes voies sont altérées dans les MICI: NOD2 (MC), autophagie et le stress du réticulum endoplasmique. Les MICI sont caractérisées par une altération de l’homéostasie intestinale associée à une augmentation d’activité des protéases dans l’intestin. Toutefois, les liens entre ces voies MICI et l’homéostasie protéolytique de l’intestin n’ont jamais été décrites. Mon projet a pour but d’étudier (1) l’impact de chacune des voies MICI sur l’homéostasie protéolytique de l’intestin et (2) l’impact des protéases sur l’activation des voies MICI. Un focus sera fait sur le « protéasome » synthétisé par les cellules épithéliales de l’intestin. Cette approche devrait apporter de nouvelles connaissances dans la physiopathologie des MICI et pourrait permettre de définir de nouvelles voies d’intervention dans les processus moléculaires des MICI.

Crohn's disease and ulcerative colitis are the two major forms of inflammatory bowel diseases (IBD). Four major pathways are altered in IBD: NOD2, autophagy and endoplasmic reticulum stress. IBD are characterized by alterations of the gut homeostasis linked with increased activity of proteases in gut mucosa. However, crosstalk between these IBD pathways and increased proteolytic activity of the gut is unknown. The aim of this project is to investigate the relationship between these IBD pathways and proteolytic activation in the gut mucosa by studying (1) the impact of these IBD pathways on the regulation of proteolytic activity of gut mucosa and (2) the impact of proteases in the induction of these IBD pathways. A particular focus will be made on the proteolytic activity of intestinal epithelial cells. This approach should provide new insights in the pathophysiology of IBD and may help to define new ways to intervene in the molecular processes involved in IBD
