Rôle des lipides bactériens dans la douleur viscérale
Le syndrome de l'intestin irritable (SII) est le trouble gastro-intestinal le plus commun. La difficulté d'identifier la cause du SII est unverrou pour le développement de thérapies. Élucider les mécanismes responsables du SII mènerait à l'identification demédicaments spécifiques et permettrait de renforcer la lutte contre ce syndrome délétère. Une dysbiose bactérienne a été décrite chez des patients atteints du SII mais son rôle n’a pas été déterminé. Nous avons démontré que les Escherichia colipeuvent synthétiser une large diversité delipopeptides et d’acides gras longues chaines en fonction de leur fond génétique. Ces lipopeptides et ces acides gras longues chaines sont capables de réguler l'activité des neurones sensitifs et la douleur. L’objectif principal de ce projet de thèse est de transférer l’étude ces lipopeptides et acides gras longues chaines à l’ensemble du microbiote en condition normale et en condition pathologiques chez les patients atteints de SII.
Role of intestinal bacteria-derived lipids in visceral pain
Irritable bowel syndromes (IBS) are the most common gastrointestinal disorders. The difficulty of identifying the cause of IBS is still amajor lock to develop effective therapies against this syndrome. Elucidating the mechanisms driving IBS disorders can lead toidentification of specific drugs and would strengthen the struggle against this highly deleterious syndrome. A dysbiosis has been described in IBS patients but its role is still unknown. We have demonstrated that Escherichia colicould synthetize a broad diversity oflipopeptides and long chain fatty acids depending on their genetic background. These lipopeptides and these long chain fatty acids are able to regulate neuronal activation and pain sensation. The major objective of this PhD project is to transfer the study of bacterial lipopeptides and long chain fatty acids to the microbiota in normal condition and in pathological conditions to IBS patients.
