Régénération de l’épithélium intestinal dans les maladies inflammatoires chroniques intestinales et les polyposes digestives
Il existe un renouvellement pathologique de l’épithélium intestinal dans les maladies inflammatoires chroniques de l’intestin (MICI) et dans la polypose adénomateuse familiale (PAF). Une hyperprolifération et un défaut de cicatrisation caractérisent respectivement les PAF et les MICI. Ce sont les cellules souches intestinales (CSI) qui contrôlent ce renouvellement rapide. Il existe 2 populations de CSI, les CBC (columnar basal cells) et les CSI situées en position +4 par rapport au fond des cryptes (CSI +4). Ces 2 types de CSI peuvent se différentier en différentes cellules épithéliales intestinales. Cependant, elles ont des fonctions métaboliques, de migration et de survie au stress différentes. L’homéostasie de la crypte intestinale est contrôlee par de nombreuses voies de signalization (Wnt, BMP, Notch, EGF, …). La voie Wnt/APC/-catenin est majeure pour le fonctionnement des CSI et altérée dans la PAF.
Nous étudions les mécanismes physiopathologiques impliqués dans ces maladies au niveau de la crypte intestinale colique.
Intestinal epithelium renewal in inflammatory bowel diseases and intestinal polyposes
IIntestinal epithelium renewal is abnormal in familial adenomatous polyposis (FAP) and inflammatory bowel diseases (IBD). An excessive proliferation or an abnormal healing is found in FAP and IBD respectively. This rapid renewal is based on the properties of intestinal stem cells (ISCs). At the bottom of the crypt are located 2 populations of ISCs: the columnar basal cells (CBC) and ISC located at the +4 position from the bottom of the crypt (ISC+4). Both ISCs can differentiate into absorptive or secretory intestinal epithelial cells. However, CBC and ISC+4 could have different metabolic, migratory functions, or stress survival. Intestinal crypt homeostasis is controlled by several pathways (Wnt, BMP, Notch, EGF, …). Wnt/APC/-catenin pathway is essential for ISCs proliferation and altered in FAP.
We study the pathophysiological mechanisms involved in these disease at the colonic crypt level
